牛血清白蛋白(Bovine Serum Albumin, BSA)是牛血清中的一種球蛋白,又稱第五組分(需要注意的是,第五組分并不是BSA的一種組份,而是Edwin J. Cohn教授早期從血液中通過(guò)特定的分離技術(shù)得到5種組份,其中第五種就是白蛋白),CAS號(hào)為9048-46-8,包含583個(gè)氨基酸殘基,分子量為 66.430 kDa,等電點(diǎn)為4.7。牛血清白蛋白在生化實(shí)驗(yàn)中有廣泛的應(yīng)用,例如在western blot中作為封閉試劑。血液中的白蛋白主要起維持滲透壓作用、PH緩沖作用、載體作用和營(yíng)養(yǎng)作用。在動(dòng)物細(xì)胞無(wú)血清培養(yǎng)中,添加白蛋白可起到生理和機(jī)械保護(hù)作用和載體作用。
低脂肪酸牛血清白蛋白產(chǎn)品及應(yīng)用
牛血清白蛋白(低脂肪酸),或者稱為無(wú)脂肪酸牛血清白蛋白,或者稱為Fat Acid Free BSA,產(chǎn)品不含有或含有極微量的脂肪酸。常規(guī)方法純化的BSA難以有效去除其結(jié)合的脂肪酸。除了特別純化的BSA產(chǎn)品外,市售的普通BSA試劑含脂肪酸量的范圍一般是: 每mol BSA含0.2~1.5 mol脂肪酸。然而,一些生物醫(yī)學(xué)研究要求使用不含游離脂肪酸的BSA,否則BSA所含高濃度的脂肪酸將影響細(xì)胞培養(yǎng)、分化、代謝反應(yīng)、生化測(cè)定等。例如,在3T3-L1脂肪細(xì)胞分化過(guò)程中不能使用普通BSA試劑,否則影響分化和細(xì)胞代謝狀態(tài);在測(cè)定培養(yǎng)的組織細(xì)胞釋放的游離脂肪酸濃度時(shí),也必須用不含脂肪酸的BSA,否則難以準(zhǔn)確測(cè)定甘油三酯水解和游離脂肪酸釋放。
BSA低脂肪酸產(chǎn)品是使用專利的熱處理方法純化而來(lái)的超純粉末。經(jīng)過(guò)專門的去脂純化處理,脂肪酸含量低于0.05% (w/w),性價(jià)比高,方便溶解。該無(wú)脂肪酸BSA可用于脂肪代謝相關(guān)分析的載體蛋白,也可與某種特異性的脂肪酸結(jié)合以形成白蛋白,可應(yīng)用于增強(qiáng)細(xì)胞培養(yǎng) 體系的性能;若實(shí)驗(yàn)中脂肪或脂肪酸的存在影響激素或者膽固醇的讀數(shù)時(shí),這個(gè)產(chǎn)品是首選;還可以作為某些脂肪以及脂肪酸分析的替代品。

General Description: Bovine
Serum Albumin (BSA) is the major protein species found in bovine blood plasma
and has applications in life science disciplines such as cell culture, in-vitro
diagnostics, human and veterinary pharmaceuticals, molecular biology, serology
and general research.
Bovogen Biologicals' BSA (Fraction V) is manufactured
using a proprietary heat shock process under GMP conditions. The BSA purity is consistently measured at 99 – 100%.
The BSA is a premium grade
product that is ideal for a variety of applications requiring a product with
high protein purity and low endotoxin. It is also fatty acid free,
IgG free, protease free and is bovine virus free.
Raw Material Origin: Manufactured from
the plasma of animals of 100% Australian origin that have undergone veterinary
ante- and post-mortem inspection and have been approved for human consumption.
